Analysis of Lymphocyte Subsets of Peripheral Blood in patient with Gastric Cancer, Using Two-Color Flow Cytometry


Abstract
To elucidate alteration of peripheral lymphocyte subsets in patients with gastric cancer, the author analyzed T cell subsets, NK cells and B cells, using two-color flow cytometry in 33 cases of stomach caner patients, 12 cases of gastic benign
disease
patients as well as 19 cases of adult healthy subjects as control.
Diagnoses were confirmed histopathologically in all the patients. All blood samples were obtained from veins of the patients and control subjects in Pusan National University Hospital during the period of January to August, 1992.
The results obtained were summarized as follow :
1. The percentage and absolute number of CD3+ CD4+ cells were significantly decreased in stage IV gastric cancer patients as compared with adult healthy controls and gastric benign disease patients, and tended to be decreased as the stage of
gastric
cancer was advanced.
2. The CD4/CD8 ratio tended to be decreased in stage IV gastric cancer patients as compared with adult healthy controls and gastric benign disease patients.
3. The percentage of CD3+ HLA-DR+ cells tended to be increased in stage III and IV gastric cancer patients compared with adult healthy controls.
4. The percentage and absolute number of Leu 3a+ Leu 8+ cells were significantly decreased or tended to be decreased in stage IV gastric cancer patients as compared with adult healthy controls and gastric benign diseases patients, and tended to
be
decreased as the stage of gastric cancer was advanced.
5. The percentage and absolute number of Leu sa+ Leu 15- cells were significantly decreased in stage III and stage IV gastric cancer patients as compared with adult healthy controls.
6. The percentage of CD3- CD16+ CD56+ cells was significantly increased in stage IV gastric cancer patients as compared with adult healthy controls and gastric benign disease patients.
On the basis of this results, it can be suggested that the impairement of cellular immunityin advanced gastric cancer patients is due to decrease of suppressor inducer T cells and cytotoxic T cells of peripheral blood lymphocyte.
